In the opening scene of the movie, Dominic "Dom" Toretto (Vin Diesel) was sentenced to jail for the crimes he conducted. As Dom was on the way to the US prison by bus, Brian O'Conner (Paul Walker) and Mia Toretto (Jordana Brewster) launched an assault on the bus, causing it to overturned and freed Dom from the bus.

While authorities hunt for them, the trio escapes to Rio de Janeiro. They went on split ways. Mia and Brian met up with an old friend, Vince (Matt Schulze) while waiting for Dom to arrive. Both of them were invited to join a job to steal three cars from a train. While on the train, Brian and Mia felt something suspicious about the job as they discover the train was carrying DEA agents and that the cars were seized property.

When Dom arrived, he realised that one of them, Zizi (Michael Irby), was particularly interested in stealing the Ford GT40. Dom asked Mia to drive the car away by herself and wait for his call while Dom and Brian fought Zizi and his companion. Soon, they caught the attention of the DEA agents which was on the train. Zizi killed the DEA agents and blamed it on Dom and Brian. Dom and Brian were captured and brought to the owner of the cars - Hernan Reyes (Joaquim de Almeida),a crime lord in Rio de Janeiro. He ordered the pair to be interrogated to discover the location of his Ford GT40. However, they managed to escape and they retreated to their safehouse.

Once at the safehouse, they examined the car to discover its importance, Vince arrived and revealed that he is working for Reyes. He removed a computer chip from the car. Dom was angry with Vince. Dom forced Vince to leave. After they investigated the chip, Brian discovered that it contain the details of Reyes' criminal empire including the locations of $100 million cash.

As Dom and Brian were blamed for the death of the DEA agents, U.S. DSS agent Luke Hobbs (Dwayne Johnson) and his team were sent to Rio to locate and capture them. They managed to locate Dom's safehouse with help of local officer Elena Neves (Elsa Pataky). However, when they arrived at the safehouse, they witnessed that they trio were being attacked by Reyes' men. Brian, Dom and Mia ran for their life and managed to escape from Reyes' men. Dom suggested that they split up and leave Rio. But Mia disagrees with the plan. Mia announced that she was pregnant with Brian's child and do not want to split from her only family member, her brother, Dom. Dom was very happy with the news and decided not to leave her sister again. Instead, he came out with a briliant plan to steal Reyes' money to start a new life.

The trio formed a team for the $100 million heist, recruiting Han Seoul-Oh (Sung Kang), Roman Pearce (Tyrese Gibson), Tej Parker (Ludacris), Gisele Yashar (Gal Gadot), Tego Leo (Tego Calderón) and Rico Santos (Don Omar). Later, Vince joined the team after saving Mia from being captured by Reyes' men. Hobbs and his team eventually found and arrested Dom, Mia, Brian and Vince.

While being transported them to an airport, they were attacked by Reyes' men. Reyes' man killed Hobbs' team and Vince. Dom, Brian and Mia fought back against Reyes' men and saved Hobbs.

Wanting revenge for his murdered team, Hobbs and Elena agreed to help with the heist. The gang rammed into the police station where Reyes' money was kept and pulled out the vault from the building using their cars. They dragged the vault all over the city with police chasing after them. Thinking that they cannot outrun the police, Dom asked Brian to continue on without him. Dom asked Brian to take good care of Mia. Dom launched an attack on the police by smashing the police cars using the vault which was attached to his car. He managed to smash Reyes' car but Zizi survived the crash. He crawled out from the car and tried to kill Dom but Brian returned and he killed Zizi. While Reyes was badly injured by Dom's assault, Hobbs arrived on the scene and shot Reyes without hesitation. Thankful for what the duo had did for him, Hobbs agreed to give them a 24-hour head start to escape but they have to leave the vault behind. Dom and Brian left in their cars.

As soon as they left, Hobbs opened up the vault only to find that it was a brand new vault with no money in it. The heist gang swapped the original vault with a new vault when they passed through a tunnel. The gang took their share and went on seperate ways. Brian and Mia escaped to a beach side paradise where Dom and Elena went to visit them. In the end, Brian challenged Dom to a final race, to prove he is the better driver.

In the post-credits scene, Hobbs was given a file by U.S. Customs agent Monica Fuentes (Eva Mendes) concerning a hijack of a military convoy in Berlin. Hobbs was only interested in news regarding Dom but Monica persuaded him to take a look at the file. The file shown a recent photo of Leticia Ortiz (Michelle Rodriguez), Dom's deceased girlfriend, revealing that she was still alive.
