After years of battle between humans and vampires, the church created a secret society, comprised of humans with super speed and strength to fight the vampires. They were known as Priests.

In a future where the war between humans and vampires was coming to an end, a team of Priests were sent to a hive to kill the Vampire Queen but it was a trap. One of the priests was taken by the vampires. As the church thought that the human had won the war, the church united the remaining human race in a city surrounded by high walls in the desert. The secret society of priests was abolished and the priests were left to go their own way.

One day, Hicks (Cam Gigandet) approached "Priest"(Paul Bettany) and told him that his girlfriend, Priest's niece Lucy (Lily Collins) was kidnapped by a group of vampires. He also added that Shannon (Madchen Amick), "Priest" ex-girlfriend was killed by the vampires and "Priest's" brother Owen (Stephen Moyer) were badly injured by the vampire.

Later, it was revealed that Black Hat (Karl urban) was the master mind behind the kidnapping. Black Hat was the member of Priest that was taken by the vampire during their last attack to the hive. Priest went to the church to ask for permission to go and search for his niece. The church disallows his will but he went against Monsignor Orelas (Christopher Plummer), the leader of the Church. He and Hicks then sets off rescue Lucy. Orelas then sent a group of four Priests led by Priestess (Maggie Q) to go after him to get him back.

When Priest and Hicks arrived at a vampire reservation, they found out that the vampires were hiding in Mira Sola, a vampire hive. Priest recalled that Mira Sola was the place where he lost his comrade which now known as Black Hat. Priestess also arrived at Mira Sola and she decided to help them. The discovered a breeding ground for the vampires. The vampires were preparing for the last attack on the human race in the city at the desert. The inhabitants of Mira Sola also dug through the mountain to the city of Jericho, where the other three Priests were heading.

Once in Jericho, the other three Priests confronted Black Hat. Black Hat then revealed himself as the Priests who were captured by the vampiressduring their last mission at Mira Sola. Black Hat was turned by the Vampire Queen into the first human-vampire. Black Hat, now equipped with faster speed and better strength, killed the three Priests as his army of vampires started a massacre at the town.

When Priest, Priestess and Hicks reached Jericho, the town was fully demolished. They discovered that the vampires were using the train tracks to travel. The next stop for the train is the city in the desert. Their only way to stop the train is to destroy the track. Hicks then found out that Priest is actually Lucy's father. They followed the train track and eventually caught up to the train. Hicks and Priest boarded the train while Priestess plants a bomb on the tracks.

After Priest and Hicks boarded the train, they were attacked by a large group of vampires. They fought the vampires and managed to escape into a compartment where they were confronted by Black Hat. Black Hat threw Hicks out of the train and then he fought with Priest. Black Hat which is much stronger than Priest defeated him and took Lucy to the top of the train. Meanwhile, Priestess defeated some of the Familiars (vampires' slaves) who tried to stop her from bombing the track.

After she killed the Familiars, she realised that one of them damaged the explosives which she had earlier placed on the tracks. Out of desperation, she strapped the bombs to her motorcycle and rode it towards the train. Just as Black Hat was about to bite Lucy, Priest came to the rescue. At the same time, he saw Priestess motorcycle raging towards the train. Just as the bike was approaching the train, he quickly hugs Lucy and jumped off the train. The motorcycle collided with the train and exploded. Black Hat was engulfed in flames, incinerating him, leaving only his hat behind. Hicks arrived and hug Lucy. Priest stood up to look for Priestess and he saw her walking towards him.

Priest then returned to the city and confronted Orelas with a vampire head. He told him that the vampires had a new army and that the war had just begun. He then walked out of the church.
